Modification of novel nanoporous (15 – 50 nm) silicagels and other silica and organic polymer based solid carriers functionalized with aminoalkyl and other modifying groups, with chelating ligands. Developing standardized processes for filling the new carriers into the biologically inert columns.
Optimization of the expression, fermentation and homogenization of the chosen recombinant target proteins: a novel thermostable lipase (selective biocatalysis, biodiesel production), a dUTPase (medical sensors), a phenylalanine ammonia-lyase (phenylketonuria, selective biocatalysis), a phenylalanine 2,3-aminomutase (stereoselective synthesis).
Development of the purification protocols for the selected recombinant proteins using the novel affinity carrier family (Variables: carrier porosity, surface modification and ligand quality, metal ion quality. Goals: capacity, purity, carrier regenerability, scalability).
Design and production of the prototypes for the controllable biocompatible pump, the thermostable test-column unit and the sterilizable X-Y-Z fluid handling robot unit, necessary for automating the purifying protocols.
Integration of the automated system. Development of the software system consisting of standard elements, responsible for the control of the system. Testing the control and the automation with the optimization of the purifying methods for the target proteins mentioned in point 3.
